目的 合成1-苯基-3-甲基-4-苯甲酰吡唑啉酮-5-苯甲酰肼腙(HL)的Pd(Ⅱ)、Pt(Ⅱ)配合物PDL2和PtL2,研究其对牛血清白蛋白(BSA)的影响。方法 利用紫外吸收光谱和静态荧光光谱定性讨论了HL以及PDL2和PtL2对BSA构象的影响。结果 HL以及PDL2和PtL2以静态猝灭的方式猝灭了BSA的内源荧光,并计算出了不同温度下这3种化合物与BSA的键合常数(Ka)以及热力学参数焓变(ΔH)及熵变(ΔS)。结论 HL和PDL2与BSA的作用力主要是静电相互作用,而对于PtL2,除了静电相互作用外还包括疏水相互作用。 |

Objective The complexes PdL2 and PtL2 were synthesized firstly by 1-phenyl-3-methyl-4-benzoyl pyrazolone-5-benzoyl hydrazine hydrazone(HL) and Pd(II), Pt(II).HL was a bidentate and its N, S atoms bond metal ions.Methods The effects of HL, PdL2 and PtL2 on the conformation of bovine serum albumin(BSA) were discussed using UV absorption spectra and static fluorescence spectroscopy.Results The results showed that they quenched the endogenous fluorescence of BSA by static quenching style.The binding constants(Ka) and the thermodynamic parameters(the enthalpy change ΔH and the entropy change ΔS) between the three compounds and BSA at different temperature were calculated.Conclusion The thermodynamic parameters suggested that electrostatic interaction force were the main force between the two compounds(HL, PdL2) and BSA.However, hydrophobic interactions were also found between PtL2 and BSA besides electrostatic interaction force. |
