      目的 分析二酰基甘油激酶Zeta(Diacylglycerol kinase Zeta,DGKZ)基因多态性(single nucleotide polymorphism, SNP)与精神分裂症(SCZ)遗传易感性的相关性。方法 211例SCZ患病者和239个健康对照者,采集外周静脉血,提取全基因组DNA,使用Taqman-MGB 探针技术检测DGKZ基因rs7951870位点多态性,对比分析两组人群等位基因分布和基因频率的差异。结果 SCZ组和对照组DGKZ rs7951870的等位基因分布和基因型频率没有显著性差异。DGKZ rs7951870:基因型χ2=2.67,P=0.26;等位基因χ2=1.07,P=0.3,优势比=0.76,95%可信区间=0.45-1.28。在加性、显性和隐性模型分析基础上rs7951870都不是SCZ的危险因素(P>0.05)。结论 本研究表明DGKZ rs7951870对中国汉族人群精神分裂症风险的影响非常小,后续仍然需要用更大样本数量的实验来验证这一发现。

      ObjectiveS This study aimed to explore the association between the single nucleotide polymorphisms (SNPs) of DGKZ and susceptibility to schizophrenia in Shandong Han population. Methods Collect peripheral venous blood and extract whole genome DNA. DNA from 239 healthy controls and 211 schizophrenic patients was genotyped for SNPs rs7951870 in DGKZ using a predesigned TaqMan SNP genotyping assay. Results Differences in genetic distributions between cases and controls were compared using the χ 2 -test. No significant differences in allelic or genotypic frequencies of DGKZ rs7951870 were detected between cases and controls (DGKZ rs7951870: χ 2 = 2.67, P = 0.26 by genotype, χ 2 = 1.07, P = 0.3, odds ratio = 0.76, 95% confidence interval =0.45-1.28 by allele. On the basis of additive, dominant and recessive model analysis, rs7951870 is not a risk factor for SCZ (P>0.05). Our results suggest that DGKZ rs7951870 SNPs are unlikely to play a major role in the pathogenesis of schizophrenia in Shandong Han population. However, these results should be validated by replication in larger and independent samples.
