      目的:建立肝纤维化小鼠模型,探究槐米提取物对肝纤维化模型小鼠的治疗作用。方法:使用胆总管结扎的方法诱导小鼠肝纤维化,将18只雄性昆明小鼠随机分为三组,分别为正常组、模型组、槐米提取物治疗组(FSI组),每组六只。造模过程中观察小鼠健康状态,采用微板法检测血清中谷草转氨酶(AST)和谷丙转氨酶(ALT),用可见分光光度法和TBA法检测血清中还原型谷胱甘肽(GSH)和丙二醛(MDA)的含量,最终进行病理切片并观察。结果:与正常组相比,模型组小鼠血清AST和ALT水平较正常组明显升高(P<0.001),抗氧化能力指标血清MDA高于正常组(P<0.0001),相应的血清GSH水平下降(P<0.01)。FSI组小鼠使用槐米提取物治疗后,血清AST(P<0.01),ALT(P≤0.05)水平较模型组下降,肝功能得到改善,血清MDA下降(P<0.01),且血清GSH(P≤0.05)提升至正常组水平。另外,模型组小鼠肝脏变性明显、粘连,表面粗糙,黄疸较为严重,FSI组小鼠肝脏无粘连,表面光滑,给药后黄疸现象消退。病理切片显示模型组肝细胞变性,坏死,肝组织炎性细胞浸润,胶原纤维增生形成假小叶,纤维化明显。FSI组增生的胶原纤维较模型组减少,相同面积下的炎性细胞个数减少,肝小叶结构破坏程度减轻。结论:槐米提取物能减轻肝脏纤维化程度,其机制与其抗炎、抗氧化、改善肝功能异常有关。

      Objective: To establish the model of hepatic fibrosis, explore the therapeutic effect of flos sophorae immaturus extract on the hepatic fibrosis model in mice. Methods: Hepatic fibrosis was caused by bile duct ligating in mice. Eighteen male Kunming mice were randomly divided into three groups: Control group, Model group and flos sophorae immaturus extract treatment group (FSI group), with six mice in each group. The Control group does nothing; Model group and FSI group were constructed by common bile duct ligation. Observing the health status of the mice, using the microplate method to measure the Aspartate transaminase (AST) and the glutamic pyruvic transaminase (ALT) in serum. The spectrophotometer method and TBA method were used to measure the glutathione (GSH) and malondialdehyde (MDA). Finally, make and observe the pathological section. Results: Compared with the Control group, the levels of serum AST and ALT in the Model group were significantly increased (P<0.001), and the antioxidant capacity index of serum MDA was higher than that in the Control group (P< 0.0001), corresponding, the GSH in serum was decreased (P<0.01). In FSI group, after the treatment Flos Sophorae Immaturus extract, the levels of serum AST (P <0.01) and ALT (P≤0.05) were significantly lower than those in Model group, the hepatic function was improved, the level of MDA in serum was significantly lower than that in Model group(P<0.01), and the level of serum GSH (P≤0.05) was improved to normal. Otherwise, the liver of mice in the model group had obvious degeneration, adhesion, rough surface and severe jaundice, while the liver of mice in the FSI group had no adhesion and smooth surface, and jaundice subsided after administration. Pathological sections showed that liver tissue of model group has degeneration and necrosis of hepatocytes, infiltration of inflammatory cells, and proliferation of collagen fibers to form false lobules, and obvious fibrosis in the model group. Compared with the model group, the proliferation of collagen fibers in the FSI group was reduced, the number of inflammatory cells was reduced in the same area, and the damage degree of hepatic lobule structure was alleviated. Conclusions: Flos Sophorae Immaturus extract can reduce the degree of hepatic fibrosis, and its mechanism is related to its anti-oxidation and improvement of abnormal liver function.
