      目的 探讨HIF-1α基因沉默对缺氧环境人下咽癌FaDu细胞增殖、迁移和侵袭的影响及其内在机制。方法 培养人下咽癌FaDu细胞,随机分为对照(常氧)组、CoCl2(采用200μmol/L CoCl2化学缺氧方法)组、CoCl2+Cki(缺氧+转染空质粒)组和CoCl2+ HIF-1αi(缺氧+转染HIF-1α siRNA)组。构建HIF-1α siRNA转染人下咽癌FaDu细胞,划痕、迁移和侵袭实验观察HIF-1α基因沉默对缺氧环境人下咽癌FaDu细胞增殖、迁移和侵袭的影响。Westernblot检测各组HIF-1α和MMP-2表达。结果 与对照组相比,CoCl2组人下咽癌FaDu细胞增殖(t=4.477,P<0.05)、迁移细胞数(t=3.814,P<0.05)和侵袭细胞数(t=3.392,P<0.05)显著增加;与CoCl2组相比,CoCl2+Cki组人下咽癌FaDu细胞增殖(t=2.081,P>0.05)、迁移细胞数(t=1.194,P>0.05)和侵袭细胞数(t=2.441,P>0.05)差异无统计学意义;与CoCl2组相比,CoCl2+ HIF-1αi组人下咽癌FaDu细胞增殖(t=7.040,P<0.01)、迁移细胞数(t=8.100,P<0.01)和侵袭细胞数(t=7.938,P<0.01)显著减小。与对照组相比,CoCl2组人下咽癌FaDu细胞MMP-2表达(t=4.424,P<0.05)显著增加;与CoCl2组相比,CoCl2+Cki组人下咽癌FaDu细胞MMP-2表达(t=2.352,P>0.05)差异无统计学意义;与CoCl2组相比,CoCl2+ HIF-1αi组人下咽癌FaDu细胞MMP-2表达(t=6.496,P<0.01)显著减小。结论HIF-1α基因沉默可抑制缺氧环境人下咽癌FaDu细胞增殖、迁移和侵袭,并降低人下咽癌FaDu细胞MMP-2表达。

      Objective To investigate the effect of HIF-1α gene silencing on proliferation, migration and invasion of human hypopharyngeal carcinoma cells in hypoxic environment and its underlying mechanism. Methods Human hypopharyngeal carcinoma FaDu cells were cultured and randomly divided into control (normal oxygen) group, CoCl2 (200μmol/L CoCl2 chemical hypoxia method) group, CoCl2+Cki (hypoxia+transfection with empty plasmid) group and CoCl2+HIF-1αi (hypoxia+transfection with HIF-1α siRNA) group. Human hypopharyngeal carcinoma FaDu cells were transfected with HIF-1α siRNA, and the effects of HIF-1α gene silencing on proliferation, migration and invasion of human hypopharyngeal carcinoma FaDu cells in hypoxic environment were observed by scratch, migration and invasion experiments. The expressions of HIF-1α and MMP-2 in each group were detected by Westernblot. Results Compared with the control group, the proliferation of human hypopharyngeal carcinoma FaDu cells (t=4.477, P<0.05) and the number of migrated cells (t=3.814, P<0.05) and the number of invasive cells (t=3.392, P<0.05) in CoCl2 group significantly increased; Compared with the CoCl2 group, the proliferation of human hypopharyngeal carcinoma FaDu cells (t=2.081, P>0.05) and the number of migrated cells (t=1.194, P>0.05) and the number of invasive cells (t=2.441,P>0.05) in the CoCl2+Cki group had no statistical significance. Compared with the CoCl2 group, the proliferation of human hypopharyngeal carcinoma FaDu cells (t=7.040, P<0.01) and the number of migrated cells (t=8.100, P<0.01) and the number of invasive cells (t=7.938, P<0.01) in the CoCl2+HIF-1αi group decreased significantly. Compared with the control group, the expression of MMP-2 of human hypopharyngeal carcinoma FaDu cells in CoCl2 group (t=4.424, P<0.05) significantly increased; Compared with CoCl2 group, the expression of MMP-2 of human hypopharyngeal carcinoma FaDu cells in CoCl2+Cki group had no statistical significance (t=2.352, P>0.05). Compared with CoCl2 group, the expression of MMP-2 of human hypopharyngeal carcinoma FaDu cells in CoCl2+HIF-1αi group (t=6.496, P<0.01) decreased significantly. Conclusion HIF-1α gene silencing can inhibit the proliferation, migration and invasion of human hypopharyngeal carcinoma FaDu cells, and reduce the expression of MMP-2 in human hypopharyngeal carcinoma FaDu cells under hypoxia.
