目的 以野生型和肝脏转基因T3斑马鱼为研究对象,探究硼砂对斑马鱼的发育毒性。方法 不同浓度的硼砂分别处理斑马鱼胚胎,每24 h观察并统计野生型斑马鱼的自主活动次数、孵化率、心率、畸形率、死亡率以及肝脏发育面积。结果 斑马鱼胚胎24 hpf,随硼砂浓度升高,自主活动次数逐渐降低,浓度0.6 mg/mL具有统计学差异(p<0.05);斑马鱼胚胎72 hpf和96 hpf,硼砂各浓度处理组斑马鱼心率均具有显著性降低(p<0.05);斑马鱼胚胎96 hpf,0.9 mg/mL硼砂处理组的畸形率显著性升高(p<0.05),96 hpf和120 hpf,0.9 mg/mL硼砂处理组的死亡率与空白对照组具有显著性差异(p<0.05)。120 hpf和144 hpf,0.9 mg/mL硼砂处理组斑马鱼幼鱼的肝脏面积显著减小(p<0.05)。结论 高浓度的硼砂对斑马鱼具有发育毒性。 |

Objective To study the developmental toxicity of borax to zebrafish of wild and liver transgenic T3(lfabp:EGFP). Methods Zebrafish embryo were treated with different concentrations of borax. The number of spontaneous movement, hatching rate, heart rate, malformation, mortality and liver area of zebrafish were observed and statistic every 24 h. Results When zebrafish eggs were fertilized 24 hours, with the concentrations of borax increased, the number of spontaneous movement gradually decreased, and there were statistical difference when the concentrations of borax reached 0.6 mg/mL (p<0.05). When zebrafish eggs were fertilized 72 and 96 hours, the heart rate in borax group significantly decreased (p<0.05). When zebrafish eggs were fertilized 96 hours, the malformation in borax group (0.9 mg/mL) significantly increased (p<0.05). When zebrafish eggs were fertilized 96 and 120 hours, the mortality in borax group (0.9 mg/mL) sharply increased (p<0.05). When zebrafish eggs were fertilized 120 and 144 hours, the liver area of zebrafish juvenile in borax group (0.9 mg/mL) significantly decreased (p<0.05). Conclusion High concentrations of borax had developmental toxicity for zebrafish. |
