      目的 探讨低温对大鼠雄性生殖功能的影响。方法 将36只雄性Wister 大鼠随机分为4组,分别为高暴露组、中暴露组、低暴露组和对照组,中暴露组、低暴露组和对照组8只,高暴露组12只(其中4只用于睾丸超微结构的观察)。对各暴露组进行为期45d的低温实验。实验结束后,对各组进行血清睾酮(T)与黄体生成素(LH)浓度的测定、精子质量分析,同时对高暴露组进行睾丸超微结构的观察。结果 对照组、低暴露组、中暴露组、高暴露组血清T浓度分别为(65.76±27.70)μg/L、(44.43±7.92)μg/L、(43.61±6.92)μg/L、(37.66±5.78)μg/L,血清LH浓度分别为(169.83±38.91)μg/L、(180.63±31.77)μg/L、(150.69±18.21)μg/L、(122.76±16.74)μg/L。与对照组比较,3个低温暴露组血清T浓度显著下降(P<0.05,P<0.01);高暴露组血清LH浓度较对照组和低暴露组显著降低(P<0.05,P<0.01)。与对照组比较,高暴露组精子计数、精子活动度及中暴露组精子活动度均显著降低(P<0.01)。与对照组比较,高暴露组、中暴露组的精子畸形率均显著增高(P <0.05, P<0.01)。高暴露组大鼠睾丸超微结构结果显示,曲细精管管腔面未发现成熟精子,支持细胞胞质内出现空泡,自噬小体增多,内含脂类物质、髓样物质及膜性结构。结论 低温可造成雄性大鼠生殖激素、精子质量的变化及睾丸超微机构的异常,并进而可能导致大鼠雄性生殖功能的下降。

      Objective To study the effects of low-temperature on male reproductive function in rats. Methods Thirty-six male Wister rats were randomly divided into four groups: high-exposure group, meddle-exposure group, low-exposure group and control group, medium-exposure group, low-exposure group and control group including 8 rats respectively, and high-exposure group including 12 rats (4 of them being used to the ultrastructure analysis). The exposure groups experienced a low-temperature test for 45 d. After the test, the serum concentrations of testosterone (T) and luteinizing hormone (LH) and sperm quality were measured in each group, and the ultrastructure of testis were analyzed in high-exposure group. Results The concentrations serum of T were (65.76±27.70) μg/L, (44.43±7.92) μg/L, (43.61±6.92) μg/L and (37.66 ±5.78) μg/L, and the serum concentrations of LH were (169.83±38.91) μg/L, (180.63±31.77) μg/L, (150.69±18.21) μg/L and (122.76 ±16.74) ug/L) ug/L in control group, low-exposure group, middle-exposure group and high-exposure group ,respectively.Compared with control group, the serum concentrations of T in the three exposure groups decreased significantly (P<0.05, P<0.01). Compared with control group and low-exposure group, the serum concentrations of LH in the high-exposure group decreased significantly (P<0.05, P<0.01) . Compared with control group, the sperm count, sperm motility and sperm motility in the high-exposure group were significantly lower than those in the medium exposure group (P<0.01).Compared with the control group, the sperm qualities were significantly lower in high-exposure group and middle-exposure group (P<0.05, P<0.01).The ultrastructure of testis in high-exposure group showed that no mature sperm was found in the lumen of somniferous tubule, vacuoles appeared in the cytoplasm of Sertoli cells, autophagic bodies increased and appeared lipid, medullary substances and membranous structure. Conclusion Low-temperature can result into changes in reproductive hormones, sperm quality and abnormalities in testicular ultrastructure in male rats, which may lead to the decline of male reproductive function in rats.
