      目的:建立乌头类药材及中成药中乌头碱类化合物含量的测定方法,为中成药质量安全评价提供依据。方法:以单因素实验对影响超声提取法萃取效率的提取溶剂、超声时间和提取温度等因素进行优化。采用高效液相色谱法测定两类中药和三类中成药中的乌头碱类物质。结果:确定了超声提取法的最佳萃取条件。根据建立的高效液相色谱法,乌头碱和新乌头碱分别在3.8-60.8μg/mL和5.6-89.6μg/mL浓度范围内与峰面积呈现良好的线性相关(r≥0.9994);RSD值均小于5.21%,加标回收率在97.53%-103.28%范围内。 结论:所建立的方法准确可靠,将其分别用于市售川乌、附子、附子理中丸、桂附地黄丸和伤湿止痛膏中乌头碱和新乌头碱含量分析,结果表明,乌头碱及新乌头碱含量处在安全范围内,符合药典标准。
英文摘要:
      Objective: To establish a method for simultaneous determination of aconitines in Aconitum and related proprietary Chinese medicine, and to provide a method for safety evaluation of proprietary Chinese medicine. Methods: The extraction solvent, ultrasonic time and extraction temperature were optimized by single factor experiment.High performance liquid chromatography (HPLC) was used to determine aconitine in two kinds of Traditional Chinese Medicines and three kinds of Chinese patent medicines. Results: The optimum extraction conditions for ultrasonic extraction were determined. According to the established HPLC method, the aconitine and mesaconitine exhibited good linearities (r≥0.9994) in the concentration range of 3.8-60.8μg/mL and 5.6-89.6μg/mL, separately; The values of RSD were both lower than 5.21%, and the spiked recoveries were in the range of 97.53%-103.28%. Conclusion: The established method is proved to be accurate and reliable. And this method has been used to evaluate the quantities of aconitine and mesaconitine in common Traditional and proprietary Chinese medicine including Chuanwu, Fuzi, Fuzilizhongwan, Guifudihuangwan and Shangshizhitonggao. The results indicate that the contents of aconitine and mesaconitine are in the safety range, which also meet the standard of Chinese Pharmacopoeia 2015.
