      目的 探讨二氢槲皮素对肾缺血-再灌注损伤模型大鼠的保护作用。方法 健康成年雄性Sprague-Dawley大鼠随机分为空白对照组(control组)、肾缺血-再灌注损伤模型组(model组)和二氢槲皮素组(DHQ组),每组6只。微板法和脲酶法检测各组大鼠血清肌酐(Scr)、尿素氮(BUN)水平,可见分光光度法和TBA法检测各组大鼠血清超氧化物歧化酶(SOD)和丙二醛(MDA)水平,并观察各组大鼠肾脏病理形态。结果 与control组相比,model组大鼠血清BUN和Scr水平明显升高(t=6.06,P<0.001;t=11.16,P<0.001),MDA水平明显升高(t=5.985,P<0.001),SOD水平明显降低(t=8.663,P<0.001)。与model相比,DHQ组血清Scr水平下降(t=6.612,P<0.001),BUN水平明显下降(t=6.903,P<0.001),MDA水平明显下降(t=5.11,P<0.001),SOD水平明显上升(t=10.66,P<0.001)。model组大鼠肾小管水肿、空泡样病变,肾小球系膜基质扩张,炎性细胞浸润,而DHQ组大鼠肾小管部分仍能保留绒毛结构,水肿面积减少,未出现空泡样病变,肾小球系膜基质扩张减轻,炎性细胞浸润减轻。结论 二氢槲皮素可以降低大鼠肾缺血-再灌注损伤导致的血清肌酐和尿素氮浓度升高,改善损伤肾小管的形态和功能,对肾缺血-再灌注损伤有一定保护作用,其机制与其抗损伤、抗氧化有关。
英文摘要:
      Objective To study the protective effect of dihydroquercetin on renal ischemia-reperfusion injury in rats and its related mechanism.Methods Healthy adult Sprague-Dawley rats were divided randomly into normal control group,model group and DHQ group.The microplate and urease method were used to measure the serum creatinine (Scr) and blood urea nitrogen (BUN),respectively.The spectrophotometer method and TBA method were used to measure the blood content of superoxide dismutase (SOD) and malondialdehyde (MDA),respectively.Results Compared with normal control group,the BUN and Scr content of model group was increased obviously (t=6.06,P<0.001;t=11.16,P<0.001),the serum MDA content was significantly higher(t=5.985,P<0.001),and the SOD content was significantly lower(t=8.663,P<0.001).Compared with the model group,Scr decreased(t=6.612,P<0.001),BUN decreased significantly(t=6.903,P<0.001),serum MDA decreased significantly(t=5.11,P<0.001),and SOD increased significantly(t=10.66,P<0.001) in DHQ group.Pathological sections showed that the renal tubules of model group rats were edematous and vacuolar like lesions,the glomerular mesangial matrix was expanded,and the inflammatory cells were infiltrated.The renal tubules of DHQ group rats still retained the villous structure,the edematous area was reduced,and there was no vacuolar like lesions,the expansion of the glomerular mesangial matrix was reduced,and the infiltrated inflammatory cells were reduced.Conclusion Dihydroquercetin can reduce the increase of serum creatinine and urea nitrogen concentration caused by renal ischemia-reperfusion injury,improve the morphology and function of injured renal tubules,and have a certain protective effect on renal ischemia-reperfusion injury.The mechanism is related to its anti-injury and antioxidation.
