      目的 探讨芒果苷对小鼠肝缺血-再灌注损伤的保护作用及其作用机制。方法 成年昆明小鼠24只,体重18~20g,随机分成缺血-再灌注损伤组(I/R组)、芒果苷组和假手术组(sham组)。I/R组采用夹闭肝蒂30min、再灌注6h制作肝缺血-再灌注损伤模型。芒果苷组于缺血前30min时腹腔注射芒果苷20mg/kg;sham组和I/R组腹腔注射等容量生理盐水。再灌注6h时,取材,HE染色,光学显微镜下观察肝细胞形态学变化;比色法检测血清丙氨酸氨基转移酶(ALT)的水平及肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的活性;Western blot检测肝组织Caspase-3、Bcl-2、Bax蛋白表达。结果 经芒果苷处理后,芒果苷组与I/R组相比肝细胞形态学异常明显减轻;血清ALT(133.73±11.53)含量和肝组织MDA(7.59±0.97)含量较I/R组明显降低,差异有统计学意义(t=-7.22,P< 0.05;t=-3.07,P< 0.05);肝组织SOD(119.40±13.95)和GSH-Px(97.59±14.07)含量较I/R组升高,差异有统计学意义(t=3.34,P< 0.05;t=3.32,P< 0.05)。经芒果苷处理后,芒果苷组与I/R组相比肝组织Caspase-3及Bax蛋白表达降低,差异有统计学意义(t=-3.18,P< 0.05;t=-8.86,P< 0.05);Bcl-2蛋白表达升高,差异有统计学意义(t=4.47,P< 0.05)。结论 芒果苷对小鼠肝缺血-再灌注损伤具有保护作用,其保护机制可能与其抗氧化作用和抑制细胞凋亡有关。

      Objective To investigate the protective effects of mangiferin on hepatic ischemia-reperfusion (I/R) injury in mice.Methods Twenty-four healthy Kunming mice,weighing 18~20g,were randomly divided into three groups:I/R,mangiferin and sham.The hepatic I/R injury model of I/R group was clamped the hepatic artery,portal vein,and common bile duct with a microvascular bulldog clamp for 30 min,and then removed the clamp and reperfused for 6h.In mangiferin group,20mg/kg mangiferin was injected intraperitoneally before 30min of ischemia.The equal volume of normal saline was given instead of mangiferin in sham and I/R groups.At 6h of reperfusion,blood samples were collected from the heart and then the hepatic tissue was taken out.Hepatic morphology of hematoxylin-eosin (HE) dying were observered by microscope.The activation of aspartate aminotransferase (AST),superoxide dismutase (SOD),malondialdehyde (MDA),and glutathione peroxidase (GSH-Px) was measured by the OD values.The expression of Caspase-3,Bcl-2 and Bax protein in the hepatic tissues were detected by Western blot.Results Mangiferin rescued hepatocyte damage by morphological observation.Mangiferin contents of ALT (135.59±19.07) and MDA (7.59±0.97) were lower than those in I/R group and the difference was statistically significant (t=-7.22,P< 0.01;t=-3.07,P< 0.01).The activities of SOD (119.40±13.95) and GSH-Px (97.59±14.07) in the hepatic tissues were higher than those in I/R group,and the difference was statistically significant (t=3.34,P< 0.01;t=3.32,P< 0.01).Mangiferin contents of Caspase-3 and Bax protein expression levels were lower than those in I/R group,and the difference was statistically significant (t=-3.18,P< 0.01;t=-8.86,P< 0.01).And the expression of Bcl-2 protein were increased than that in I/R group,and the difference was statistically significant (t=4.47,P< 0.01).Conclusion Mangiferin can significantly protect the hepatic tissue against the I/R induced injury.The mechanism may be related to the effect of anti-oxidant and inhibiting cell apoptosis.
