摘要 目的 探讨锌指蛋白GLIS3在肝细胞肝癌(HCC)中的表达及其临床意义。方法 收集TCGA、GTEx和GEO等数据库中GLIS3在肝细胞肝癌中的表达值,对2101例HCC组织和1448例正常肝组织进行综合分析,用实时定量逆转录聚合酶链反应技术验证在肝细胞癌细胞系(MHCC-97H、SK-HEP-1)和正常肝细胞细胞系(LX2)中GLIS3的表达,免疫组化技术验证96例肝细胞癌组织和96例癌旁组织GLIS3的表达水平。结果 数据库分析结果显示,GLIS3在癌组织中的表达明显低于非癌组织(SMD=-0.50;95%CI=-0.69-0.31;I2=80%;P<0.001)。实验验证GLIS3在MHCC-97H、SK-HEP-1肝细胞癌细胞系中的表达明显低于正常肝细胞细胞系(P<0.0001)。GLIS3蛋白的差异表达与HCC患者肿瘤数目相关(P<0.05)。结论 GLIS3在肝细胞肝癌组织中表达较低。可能成为肝癌治疗潜在的靶点、诊断和预后判断的标记物。 |

Abstract Objective To investigate the expression and its clinical significance of the zinc finger protein GLIS3 in hepatocellular carcinoma (HCC) . Methods The expression levels of GLIS3 in hepatocellular carcinoma were collected from databases such as TCGA, GTEx, and GEO, and a comprehensive analysis was conducted on 2101 HCC tissues and 1448 normal liver tissues. Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) was utilized to validate the expression of GLIS3 in hepatocellular carcinoma cell lines (MHCC-97H, SK-HEP-1) and a normal liver cell line (LX2). Immunohistochemistry was used to verify the expression levels of GLIS3 in 96 hepatocellular carcinoma tissues and 96 adjacent non-tumor tissues. Results Database analysis indicated that the expression of GLIS3 was significantly lower in cancer tissues compared to non-cancer tissues (SMD = -0.50; 95% CI = -0.69 to -0.31; I2 = 80%; P < 0.001). Experimental validation confirmed that the expression of GLIS3 in MHCC-97H and SK-HEP-1 hepatocellular carcinoma cell lines was significantly lower than that in the normal liver cell line (P<0.0001). The differential expression of GLIS3 protein was related to the number of tumors in HCC patients (P < 0.05). Conclusion GLIS3 is expressed at lower levels in hepatocellular carcinoma tissues. It may serve as a potential target for liver cancer treatment and a biomarker for diagnosis and prognosis. |
