      目的 探讨采用脂肪乳剂、30%白酒灌胃和皮下注射CCl4相结合建立SD大鼠脂肪肝动物模型的方法。 方法 SD大鼠随机分为正常对照组与模型组,正常对照组普通饲料喂养,模型组喂养普通饲料并给予脂肪乳剂、30%白酒灌胃,皮下注射CCl4,连续4周。动态观察一般情况及体重变化,HE染色观察肝脏病理动态变化过程,4周末计算肝指数,检测各组大鼠生化血清指标及肝组织中TC、TG、FFA含量。 结果 模型组大鼠体重低于正常对照组(P < 0.05)、肝指数高于正常对照组(P < 0.05);镜下观察模型组大鼠肝脏脂变程度已达1/3,符合FL的特征;与正常对照组相比,模型组大鼠血清ALT、AST活性升高(P < 0.001);LDL、TC、TG含量升高(P < 0.001),HDL降低(P < 0.001),FFA升高(P < 0.001);模型组大鼠肝组织TC、TG、FFA含量高于正常对照组(P < 0.001)。 结论 通过4周脂肪乳剂、30%白酒灌胃与皮下注射CCl4相结合的复合造模方法可成功制备SD大鼠脂肪肝动物模型。

      Objective To establish fatty liver (FL) model in SD rats by fat emulsion, intragastric administration of 30% alcohol and subcutaneous injection of CCl4. Methods SD rats were randomly divided into normal control group and model group.The normal control group was fed with ordinary fodder, while the model group was fed with ordinary fodder, together with fat emulsion and 30% alcohol, and subcutaneous injection of CCl4 for 4 weeks.The general situation and weight changes were dynamically observed.The dynamic changes of liver pathology were observed by HE staining.At the end of 4 weeks, the liver index was calculated, the biochemical serum indexes and the contents of TC, TG, FFA in liver tissue were measured. Results The body weight of the model group was significantly lower than that of the normal control group (P < 0.05), the liver index was significantly higher than that of the normal control group (P < 0.05).Microscopically, the degree of hepatic steatosis in the model group has reached 1/3, which is consistent with the characteristics of FL.Compared with the normal control group, the activities of serum ALT and AST in the model group were significantly higher (P < 0.001);the contents of LDL, TC and TG increased significantly (P < 0.001), HDL decreased significantly (P < 0.001), FFA increased significantly (P < 0.05).The contents of TC, TG and FFA in liver tissue of model group were significantly higher than those of normal control group (P < 0.001). Conclusion The animal model of fatty liver in SD rats can be successfully established by the combination of fat emulsion, 30% alcohol gavage and subcutaneous injection of CCl4 for 4 weeks.
