目的 探讨Apelin-36对神经细胞内钙离子超载导致的内质网应激的抑制作用。方法 将Wistar大鼠随机分为假手术组和模型组,每组6只,RT-PCR法检测两组大鼠皮层脑组织CHOP、GRP78mRNA的表达;利用SH-SY5Y细胞构建钙离子超载细胞模型,RT-PCR法检测Apelin-36对CHOP、GRP78mRNA表达的影响。结果 与假手术组相比,模型组大鼠CHOP和GRP78mRNA表达显著增加,差异具有统计学意义(t=5.832,P<0.05;t=5.938,P<0.05)。在SH-SY5Y细胞中,与对照组比较,细胞模型组CHOP和GRP78mRNA表达显著增加,差异具有统计学意义(均P<0.05)。与细胞模型组相比,Apelin-36组CHOP和GRP78mRNA表达显著降低,差异具有统计学意义(均P<0.05)。结论 Apelin-36可能通过抑制钙离子超载导致的内质网应激实现对脑卒中神经细胞的保护作用,为Apelin-36应用于临床治疗提供了理论基础。 |

Objective To investigate the inhibitory effect of Apelin-36 on endoplasmic reticulum stress induced by calcium overload in neurons.Methods Wistar rats were randomly divided into sham operation group and model group.The expression of CHOP and GRP78 mRNA in the cortical brain tissue of two groups were detected by RT-PCR.The calcium overload cell model was constructed by SH-SY5Y cells.The effect of Apelin-36 on the expression of CHOP and GRP78 mRNA was detected by RT-PCR.Results Compared with the sham group,the expression of CHOP and GRP78 mRNA in the model group were significantly increased(t=5.832,P<0.05;t=5.938,P<0.05).Compared with the control group,the expression of CHOP and GRP78 mRNA in SH-SY5Y cells were significantly higher than those in the control group(P<0.05).Compared with the cell model group,the expression of CHOP and GRP78 mRNA in Apelin-36 group was significantly lower than that in the control group(P<0.05).Conclusion Apelin-36 may protect the neurons of stroke by inhibiting the stress of endoplasmic reticulum induced by calcium overload,which provides a theoretical basis for the application of Apelin-36 in clinical treatment. |
